Very proud of this collaborative work, where combination of expertise brought us one step further (and made it more fun)! I find particularly exciting that AIS and synaptic plasticity co-occur and share common molecular mechanisms. Clearly, this will be followed up!

We tried (veryyyyyy hard) to visualize internalization of Nav1.2 channels during AIS plasticity. What we saw is the appearance of Nav1.2 vesicles colocalizing with dynamin2, and an increase in endocytic structures at the distal AIS during plasticity in the presence of Dynasore.

@mariescopy @HenriquesLab Moved to the NL with my 1.5yo, first 3 months have been frustrating for him, but now he's fluent in Dutch! Going to a bilingual or international school might help, because teachers are aware of language issues/integration and kids are used to switch between several languages!

In short, 1. We reveal a new function for 480AnkyrinG. Once targeted to the membrane, 480AnkyrinG efficiently recruits TRIM46-decorated microtubules, in an EB-dependent manner.

We first developed knock-ins to label endogenous Nav1 channels, and used the AnkyrinG-GFP mouse line generated by @jenkins_paul and @AxonalLab . We were excited, because these tools could allow us to follow endogenous AIS major proteins during plasticity.

To image the AIS of individual neurons during plasticity, we used endogenous labeling of (i) AnkyrinG using the cool mouse line shared by @AxonalLab and (ii) Nav1.2 channels using the ORANGE toolbox developed by @Jelmerwillems and @hdmacgillavry .

I find really exciting that plasticity at the AIS and synapses occurs in parallel and shares common molecular pathways. More work has to be done to fully understand the co-operation between synaptic and intrinsic plasticity and its functional consequences for network function!

3. TRIM46-mediated microtubule organization is crucial for the proper trafficking to the proximal axon of other AIS proteins, such as Neurofascin-186. Once in the proximal axon, AnkyrinG allows for the stable accumulation of Neurofascin by locally preventing its endocytosis.

2. We show that TRIM46 only binds to parallel microtubules and acts as a strong rescue factor. This way, it confers uniform orientation and stability to the microtubules recruited at the membrane by AnkyrinG.
