I knew very little about B cells but now my whiteboard is filled with Blagoje’s drawings. His passion is inspiring and this position is an awesome opportunity, don’t miss!
1/5. By looking at nascent RNA in different ways, we find that the length of poly(A) tails at synthesis has a broad distribution peaking at 200-250 nt, much longer than that at steady state.

From my side, really happy to see how FLAM-seq combined with Iso-seq can massively help annotating the transcriptome of non-model species. Let's do this again :)

I would probably infect iPSCs and generate cerebral organoids. Readout would be scRNAseq to infer cell types that are expanded or depleted in response to a certain TF over expression or KD. A sort of functional lineage tracing
We couple our full-length cDNA method with
@PacBio
sequencing and get high quality sequences for thousands of mRNAs in cell lines, organoids and C. elegans.
We discovered that poly(A) tail length is broadly dependent on 3'UTR and also on promoter choice. Moreover, we see quite some cytosines within the tails!

5/10. We sequenced HeLa, iPSCs, brain organoids, C. elegans. Each biological sample has its peculiar poly(A) length distribution, and tail length varies from gene to gene, within approx. 25 and 250 nt. 50 nt seems a special length: all mRNAs have a peak around there.
