I believe we provide some interesting evidences on how extracellular cues target the nucleus to effect its fate and function in 2 ways. Nuclear shape and LaminA conformation rely on cytoscheletal action; chromatin fluctuations depend on two key second messengers:Ca2+and cAMP -9-

Importantly cAMP inhibition not only prevents the chromatin fluctuations but it affects the differentiation potential of the cells towards their Osteogenic commitment. -8-

We observed a strong fluctuation also of the enhancer histone mark H3K27ac. Interestingly sonochallenge induces an enlargements of RNAPII-S5p nuclear foci like Coilin despite the fact that size and numbers of the foci do not seem coincide along the timecourse. -4-
