Perspective. Uncovering previously hidden patterns in the genome and understanding how chromosomes communicate in healthy and disease genomes is next. Big THANKS to the incredible team, to @ericjoycelab, Samin Aref, Thomas Liehr, Anja Weise, and to funding agencies! @CIHR_IG.

New rules of genome topology. 61 ‘topological anchors’ occur across cell types at the speckles, where especially p- and q-telomeric contacts coalesce regions of highest gene density and transcription factor binding and synergistically shape genome topology by a spatial gradient.

Contradictory findings. Rabl’s configuration with extensive inter-telomeric and inter-centromeric clustering exists across human cell types. Despite recent findings, Rabl’s configuration and chromosomal territories may not be mutually exclusive.

Fundamental Properties. Recurrent inter-chromosomal contacts occur in gene-dense and highly expressed genomic regions that harbor cell-specific and sex-specific genes. Check your cell type of interest in Supplementary Data 3 & 4.

What are the functional roles of 3D genome organization? Our paper in @Nature led by @_prashantbhat demonstrates how dynamic 3D genome organization around nuclear speckles plays a crucial role in regulating mRNA splicing efficiency.
