Together with the Cravatt group, we have a new manuscript out reporting the chemoproteomic discovery of covalent FOXA1 ligands that bind its DNA-binding domain, corrupt its motif preferences, and redistribute it on the genome. 1/11

Best practices statements are helpful for promoting reliable technologies across different and multidisciplinary disciplines. The probe development field expanded dramatically over the past couple decades and recommended conditions/controls may help increase their use.

CONSENSUS STATEMENT. Reactive oxygen species are complex & difficult to measure. A group led by Murphy & Halliwell (@Cambridge_Uni, @NUSingapore) proposes guidelines on ROS nomenclature & assessment in cells, tissues & in vivo.

Nice reminder from @INSTMohali that sensors can alter biology. NO sensor (Urea-eTCBD) is anti-inflammatory at low μM concentrations. Additional evidence to support NONOate over N-nitroso formation (e.g., 15N NMR) and whether the adducts can release NO would be interesting.

Excited to share new paper @CellCellPress. Led by Zhengyuan Pang @ScrippsGradPrgm, collaboration w/ Ben Cravatt, CATCH is a chem-bio innovation to allow imaging of drugs in tissue. This "mission impossible" only happens with the incredibly collaborative @scrippsresearch . 1/3.

The concentration of truncated dye could then be used to report on the local environment of the cells during phototruncation and read out on downstream phenotypic changes. Congratulations @SchnermannLab, @sid_matikonda, @LenkaStackova!.

I am curious about the O2-dependence of phototruncation, particularly in the context of cancer. If the rate of phototruncation is sensitive to (patho)physiological O2 levels, then the yield may differ between hypoxic/normoxic tissues.

Interesting strategy to track cells in live animals by leveraging light-induced cyanine phototruncation. This represents a nice example of applying mechanistic understanding ( to develop a tailored dye platform with exciting applications.

Interested in detecting nitric oxide (NO) in live cells or animals with bioluminescence imaging? Consider using BL660-NO, an NO-gated wild-type firefly luciferase substrate. BL660-NO was then used to evaluate the effect of diet on NO in a murine cancer model. 🔦🧫🐭🍔@ChanLabUIUC.

Interesting work from Natasha Vetter and @DavePalmerUSask. The authors use C-phosphonate substrate analogs and phosphite rescue experiments to dissect the role of dianion activation in kanosamine biocatalysis. Brings back wonderful memories from my time in @jprichard53's lab.

Beautiful example of controlling biology with light from @DirkTrauner's group. I am curious whether this strategy can completely block dark engagement or if protein flexibility allows the enzyme to adopt a competent conformation for Cys reactivity, albeit at slower rates.
