Thanks for the nice summary. Exciting indeed to see our chemical proteomics and @Cyclica/@thesgconline's computational/structural methods converge on discovery of same druggable pocket in DCAF1.
Researchers at @scrippsresearch published work where they confirmed DCAF1 as a validated PROTAC E3-ligase. DCAF1 was the basis of @Cyclica’s work with @thesgconline in 2021 where we published the first co-crystal structure with DCAF1 bound to CYCA-117-70!.

We describe a functional screening strategy to discover bifunctional compounds that degrade proteins by covalently engaging E3 ligases, which leads to the discovery of DCAF11 as a target of electrophilic PROTACs that supports targeted protein degradation.
