We hypothesise that HAT binding may be the primary intracellular role of pro-IL-1α, and the NLS may serve an auxiliary role – either to facilitate binding to HATs in the nucleus, or instead it may be related to IL-1α trafficking and release

Interestingly, species that have lost conservation of the IL-1α NLS have maintained conservation of the HAT-binding domains. Monotremes also have evidence of an intact NLS in IL-1α, suggesting that the NLS evolved in the proto-mammal, prior to mammalian divergence.

By analysing evolutionary evidence, we found that in addition to toothed whales ( https://t.co/68kZhrW33L), other mammals such as castorimorpha (e.g. beavers) and marsupials have lost IL-1α NLS conservation. We validated that these mutations cause loss of nuclear localisation

Using proximity labelling, we identified proteins within close proximity to pro-IL-1α, including many nuclear proteins and specifically various HAT proteins, such as EP300. These data suggested that interaction with nuclear HATs could be an intracellular role for IL-1α

IL-1α has a highly conserved nuclear localisation sequence (NLS) in the pro-domain, which facilitates trafficking of pro-IL-1α to the nucleus, and two HAT-binding domains. Why does IL-1α have an NLS, and does IL-1α have a role in the nucleus?
