Excited to share our new paper describing the structure of an SpCas9 adenosine base editor (ABE) out now @ScienceMagazine! Co-first authored with @Sarkele1, we captured a base-editor in the act using cryo-EM! https://t.co/2r5VlG9OKp (1/5).

These are challenging targets, CRISPR which are large multi domain RNA binding proteins and ChuA - an uncharacterized transmembrane protein. End-to-end design and validation would not have been possible without the hard work of @CyntiaTaveneau @_danielfox and all co-authors!

We are excited to share pre-prints describing a successful foray into ML-driven protein design. With @Rhys__G we designed proteins to potently inhibit CRISPR-Cas13 ( https://t.co/QzdlK7zJ0W) or block heme piracy by pathogenic E. coli ( https://t.co/pDC6IEVDWp):

Ever wondered where Cas9 came from? @Sarkele1 and I wrote a 'First Cut' for @CRISPRjournal describing the epic work by @altaetran and colleagues on the origins of Cas9. Our summary: https://t.co/yqiYBDZej7 The epic paper: https://t.co/rlYiNkzdED Phage be like👇

Curious to know how the genome editor Cas9 unwinds DNA to "read" it? Check out this great story by @JoshCofsky from the @doudna_lab. When we saw the first cryo-EM maps Josh said: "WOW the DNA is like REALLY bent. This is what comes to mind when I'm thinking about the mechanism:"
New pre-print! While #CRISPR-Cas9 searches through the genome, how does it actually "read" DNA sequence information, which is normally hidden within the double helix? We determined structures of Cas9 in the act of DNA interrogation to find out! Thread below 👇 (1/8)
