Our most recent Nature Cell Biology paper describes an innovative Perturbation-response Score (PS) method to decode heterogeneous responses of cells to perturbations at single-cell level. Particularly helpful for analyzing Perturb-seq data.

We tested our model in various settings including T cell stimulation, pancreatic differentiation, HIV expression, dosage response of essential genes etc. In particular, @MylliaBio provides a genome-scale Perturb-seq to trackle T cell stimulation (>500k single cells).

PS enables studying cell-type specific gene functions. We tested PS in multiple Perturb-seq/scRNA-seq datasets in HIV latency and pancreatic differentiation. In particular, we discovered (and validated) a novel function of CCDC6 during pancreatic/liver differentiation (5/n).

We further applied dosage analysis of perturbations using PS, without the need to adjust perturbation strength, which is not easily scalable. For example, "sensitive" genes lead to strong expression responses even with moderate exp reduction (4/n)
