Overall, we propose that transcriptional memory mechanism is conceptually based on: . - accelerator (retention of specific active histone marks).- brake (local loss of repressive Polycomb mark).- clutch (cis-regulatory element controlling memory output). 🧵(7/8).

Since the preprint, we found that the establishment of the memory is widespread in cell population irrespective of expression status of reporter memory gene and added clustering of epigenomic features at memory genes with unsupervised learning. 🧵(6/8).

We also found that gene hyper-activation during IFNγ restimulation is repressed by a cis-regulatory silencer. This silencer forms long-range chromatin loops, which reset post-stimulation and are not epigenetically maintained. 🧵(5/8).

We found it’s driven by:.- Post-stimulus retention of active histone marks (H3K4me1, H3K14ac, H4K16ac).- Local loss of repressive Polycomb mark (H3K27me3). Strangely, H3K27me3 initially accumulates during first IFNγ stimulation when genes are activated. 🧵(4/8).
