Overall, this work offers critical insights into molecular mechanisms underlying ASD, and describes a powerful platform that combines wt and mutant PPI data with AF2 to develop and test therapeutic strategies for a wide range of genetically-defined conditions.

We observed a patient-derived mutation in FOXP1 disrupting interaction with FOXP4, leading to a change in DNA binding sites and disruption of neurogenesis in human forebrain organoids. Consistent with previous work this has an impact on deep layer cortical and subplate neurons.

Using AF2, we provide molecular-level detail of the interaction interfaces of ADS-PPIs, enabling mechanistic hypotheses on mutational impact. We observed interface mutations more often result in loss rather than gain of interaction, even for low-scoring AF2 predictions.

We also generated a differential PPI map of 54 patient-derived missense mutations. This network suggests that damaging hcASD variants share changes in molecular function, with some interactors being commonly gained or lost across multiple mutant hcASD.
