(6/7) Finally, we made a DM1 cell model from patient-derived DNA and showed that we can detect repeat-containing mRNA directly from total RNA. This opens the door to understanding the biology of repeat RNA its processing, localization, and ASO therapeutic targeting.

(5/7) We also applied it to the PHOX2B gene, where expansion of just a few tandem repeats causes congenital central hypoventilation syndrome-1 (CCHS-1). We can distinguish between as few as six tandem repeats!.

(4/7) We tested this approach on myotonic dystrophy type I (DM1), a classic repeat expansion disorder. With our system, we can detect who has DM1 from RNA by identifying the number of CTG repeats, diagnostically relevant, and enzyme-free.

(3/7) Our solution? Ditch the enzymes entirely. Using solid-state nanopores, we directly analyze molecules without unfolding or copying them. That means no polymerase, no helicase, just the molecule and a nanopore. Structure formation? Not a problem with or RNA nanostructures!.

(2/7) Enzymes like polymerases and helicases stall or fall off when faced with long repeats. Even nanopore helicase-based systems get jammed. The repeats form strong secondary structures, hairpins, quadruplexes that stop sequencing in its tracks.

Check out this tour de force study by Sangwoo Park and his incredible team of collaborators. Biophysical properties of single nucleosomes are responsible for A and B compartments in cell nucleus.
