We tried so many different things to try to optimize this platform, and learned a lot of things along the way in regards to the difficulties in sample preparation for cellular cryo-ET (with special attention to neurons).

But we encountered a LOT of difficulties along the way, with the biggest problem being that even though the neurons looked fine when grown on grids before freezing, when we put them in the EM they look like anything but.

This project started as an attempt to establish an alternative primary neuronal cell culture model for cryo-ET, using Drosophila due to the versatility in genetic manipulation, low cost of maintenance, and fast reproduction rate.

While I wasn’t able to attend the last day cause I got ill, I just wanted to say it was a blast attending my first ever SfN conference here at DC! It was great seeing lectures where cryo-EM and cryo-ET got a shoutout, and meeting so many new people and old friends too! #SfN2023
