Domains are important! Different functions & propensity for disease. Hope this helps 1. lay groundwork to distinguish cellular 'neighborhoods' 2. precisely track regional adaptations in response to diet, disease+ & 3. one day,✨regenerate specialized function in damaged guts✨8/.

So, 53 feet of tissue & 315 scRNAseq samples later, the duodenum, jejunum, & ileum, once useful, are now a thing of the past. I’m excited to provide concrete molecular framework for 5 intestinal domains. Track ‘em in tissue or in silico w/our domain identity-trained classifier 7/.

How are domains controlled? We predicted the factors that pattern them and made CRISPR-modified regional organoids to modulate them as a start to answering that question. Also, we find the positions of 3 regional stem cell populations that likely seed them 6/.

So how many regions are there? 3? 30? After extensive analysis, we found 5 distinct domains. Domains = intestinal regions inhabited by enterocytes that co-express genes that encode distinct aspects of nutrient absorption, and have individual responses to 🥯🍗🍕🍤 5/.
