It is also exciting to see that the Chem-Seq works so well with HDAC1/2 with @Nilay_Sethi and @LabSethi after the development on Chem-Seq of BET bromodomain @jaybradner.
Aberrations in epigenetic regulation prevent cellular differentiation and endow plasticity to cancer. By screening drugs using a new reporter system, we found that inhibiting HDAC1/2 promotes colorectal cancer (CRC) differentiation and anti-tumor activity.

so excited to learn more about different domain functions in different type of cancers. It is exciting for me and my team to learn more and more about pediatric brain tumor and its dependencies. @RousselMartine @ADurbinLab @Abraham_Abrahub.
But what are the effects of targeting different domains? With support from @TheAbraLab, we tested these different inhibitors in #medulloblastoma cells. In #medulloblastoma, targeting the bromodomain of EP300/CBP caused rapid loss of a network of genes required for survival.

It is so great to see that different cancers need EP300/CBP functions differently. The chemical probes can help us to identify the most efficient way to target EP300/CBP in different cancers, as MYCN-amplified neuroblastoma more relies on EP300 structural functions @CD_AACR.
Led by @NohaShendy, Melissa Bikowitz and Logan Sigua, and with Martine Roussel's lab, we were struck that #medulloblastoma cells seemed preferentially sensitive to @inobrodib. But inobrodib/CCS1477 didn't cross the blood-brain barrier, which limits its use in brain tumors.
