How is the protein NINJ1 regulated so that cells break open only when they die?

Obviously, we wont survive with NINJ1 constantly poking holes in our cells. So how is NINJ1 kept on a leash? The answer turned out to be quite simple. NINJ1 autoinhibits in trans by forming a face-to-face dimer. This shields its hydrophilic face (resembling a partner dance) (3/3)

NINJ1, a tiny membrane protein, ruptures the plasma membrane to amplify the immune response during lytic cell death. Previous papers showed active NINJ1 forms filaments (resembling a river dance) and NINJ1 filaments rupture membranes via an exposed hydrophilic face (2/3).
