All it took was moving to the east coast then two weeks later waking up at 4 am to drive 2.5 hours then take a train 1.5 hrs then the subway and a tram and waiting in line for 2.5 hours then trekking back home (by myself) and also this was my first time in NYC 😂

I was able to develop the libraries and ship them out to Berkeley where I spent 6 months in the @DeutschbauerLab optimizing and performing fitness assays and analyzing data with help from Adam Deutschbauer @HansCarlson3 & Morgan Price!

Thus coming into the 3rd year of my PhD I reached out to Adam @DeutschbauerLab and proposed applying for the SCGSR and developing the RB-TnSeq library in Methanococcus and working with him to run some fitness assays. I was awarded the SCGSR & some fitness assays became many….

I started graduate school then and I missed having this resource in a related species since working with Methanococcus, especially since COVID hit right after finishing rotations and data analysis and reading were the options available

A note on my motivation for this project - When I was an undergrad I would often browse Desulfovibrio genome info and look at RB-Tnseq data for homologs of genes of interest in other bacterial species on the FitnessBrowser from the @arkin_lab @DeutschbauerLab. So helpful!

I hope that this large dataset can be a useful tool for the archaeal community & inspire others to generate other RB-TnSeq libraries in their favorite microbe (even if it’s a strict anaerobe, that likes high concentrations of H2 and sulfide it’s possible!) 🥳

When looking at sulfite stress we found that 0.2 mM sulfite can actually be used as the sole S source if you over-express the gene encoding dissimilarity sulfite reductase-like protein (Dsr-LP)! Despite lacking the F420 oxidase domain found in Fsr, it’s functional!🤯

To my surprise, I found temporary oxygen exposure isn’t so bad when sulfide isn’t around to be oxidized to sulfite. Although there are a few genes affected
