Indeed, clearly supporting the idea that NuA4/TIP60 does not use the same molecular mechanism as SRCAP to engage nucleosomes in order to incorporate H2A.Z. The multiplicity of histone mark reader modules in NuA4 likely playing a role.
Please also take a look at related work by @rklouder and Carl Wu at @HopkinsMedicine diving into different functional states of the human SRCAP remodeler and a comparison with TIP60. Recently out in @biorxivpreprint .

So happy this is finally out. Fantastic collaboration with @NogalesLab to obtain high resolution structure of the human NuA4/TIP60 complex and investigate the function of the TRRAP subunit in recruitment.

The human nucleosome acetyltransferase of histone H4 (NuA4)/Tat-interactive protein, 60 kilodalton (TIP60) coactivator complex, a fusion of the yeast switch/sucrose nonfermentable related 1 (SWR1)...
In humans, the essential TIP60 complex is a fusion of 2 molecular assemblies that in yeast perform different functions: .-NuA4 ➡️ a histone acetyltransferase. -SWR1 ➡️ a chromatin remodeler that incorporates H2A.Z in promoter and enhancer-proximal nucleosomes. 🛠️🧬.
