After failure of ASO trials targeting C9orf72 sense repeats also targeting antisense repeats is now vital. Our new preprint shows a potential approach using CRISPR-CasRx @UKDRI @UCLIoN @AlexCammack

The most common genetic cause of both frontotemporal dementia (FTD) and amyotrophic lateral sclerosis (ALS) is a G4C2 repeat expansion in intron 1 of the C9orf72 gene. This repeat expansion undergoes...

Taken together this suggests that getting PUFAs to neurons is important for their beneficial effect. Interventions that can boost neuronal lipid desaturation could therefore provide benefit in FTD/ALS 9/9.

Feeding C9 flies PUFAs was beneficial but neuronally over-expressing fatty acid desaturase enzymes was much more effective, and the same fatty acid desaturase also reduced stressor-induced toxicity in C9 iPSC-neurons 8/9

To determine whether this deficit was relevant to neurodegeneration or just a bystander, we took two approaches: feeding C9 flies PUFAs, or over-expressing fatty acid desaturase enzymes only in adult neurons 7/9.

The signature could be prevented by treating with a C9 ASO and recapitulated by expressing repeats in control lines, showing these lipid changes are driven by the repeat expansion 6/9.

Remarkably, we saw the same lipidomic signature: a specific decrease in phospholipids containing polyunsaturated fatty acids (PUFAs) 5/9

To investigate lipid changes we performed lipidomics with @MartinGiera and @rikvanderkant on C9 iPSC-neurons and FTLD brains 4/9.

The work started when we found that the very same genes involved in lipid and fatty acid synthesis and desaturation were downregulated in a fly model of C9orf72 FTD/ALS and ALS post-mortem spinal cord. 3/9
