5/ This work reveals a metastatic transcriptional program in a therapy-naÏve context, offering insights into the biology of de novo metastatic prostate cancer and opening paths to better stratification and management.

4/ Our bioinformatic analyses point to SOX11 gene as a key regulator of this program. We validated its role both in vitro and in vivo.

3/ Using single-cell RNA-seq in two additional cohorts, we mapped this ECM signature to specific cancer cell clusters. These cells engage in aberrant heterotypic interactions with the tumor microenvironment.

2/ We performed RNA-Seq and network analyses on two independent patient cohorts. We uncovered consistent dysregulation of extracellular matrix (ECM)-related programs in these tumors.

1/ Less than 10% of prostate cancer patients are diagnosed with metastatic disease at diagnosis, but they account for 50% of prostate cancer mortality. Surprisingly, these primary tumors remain poorly characterized at the molecular level.
