When the variants of concern became prevalent, we noticed mutations around our binding site, so probed them with uSTA: sugar binding vanished!

A crack team at the @RosFrankInst got a super high-resolution structure of spike with sialic acid (the sugar of interest) bound to the NTD. Comparison of cryo-EM and NMR poses gave excellent agreement.

uSTA is easily scalable, so we studied a panel of natural/unnatural sugars. These showed an ‘end-on’ binding pose for sialoside ligands in spike, which was remarkably similar to the influenza HA pose. But how did we confirm our new method was accurate?
