Deciding which patients can receive #CAR T therapy is an important clinical question. By uncovering the structural basis of #MHC antigen recognition by a #neuroblastoma-targeting CAR we double the patient cohort and limit off-target toxicity @SciImmunology https://t.co/e0ZHaBoRFM

By solving the crystal structure of the 10LH-PHOX2B-HLA-A*24:02-β2m complex, we identify critical interactions b/w the CAR + pHLA & validate them w/ alanine scanning + SPR binding studies. The key residues are the peptide P6 arginine & framework residues along the HLA α1 helix.
