We show in vitro and in vivo that EL likely “collaborates” with LPL to promote efficient TG rich lipoprotein breakdown, possibly through phospholipid and TG lipolysis of PUFAs which allows more TG lipolysis by the predominant TG lipase LPL (6/n).

Our study shows that EL inactivation in mice and humans delays breakdown of polyunsaturated fatty acids on TG rich lipoproteins (5/n).

We further show in mice lacking endothelial lipase and biochemical studies in vitro that EL is a key contributor to TG rich lipoprotein catabolism through TG lipolysis (4/n).

Endothelial lipase (EL) is a key regulator of HDL through breakdown of phospholipids. Prior work from from @danrader333 @CDG_MDPhD @skathire @bvoight28 and many others have shown that genetic loss of LIPG/EL is a key regulator of HDL-C in humans (2/n).

Fun to work on this editorial piece on an important question that has come up often as a fellow with the marvelous mentorship of @pnatarajanmd and @mchonig. Many gaps in how we assess risk of ASCVD that don’t fall cleanly in primary vs secondary buckets and impact therapy.
