             Very nice Guille, so whatโs next? We hope to have demonstrated the potential of C. acnes to become the best microbial chassis for synbio applications in the skin and we are working to validate this in more realistic models so stray tuned!

             For therapeutics, we implemented a UV-damage kreatinocyte model and see if the supernatant of an engineered C. acnes producing a recombinant antioxidant protein could protect the cells. Turns out it reduced significantly the ROS levels in the UV exposed keratinocytes!

             For biocontainment, we focused on targeting key metabolic genes so C. acnes would only survive if we topically supplement the missing amino acid. Since knocking out genes is still challenging in this skin non-model bacterium, we turned to #CRISPRi to quickly screen auxotrophs.

             How did we apply these tools? We focused mainly on two objectives: 1๏ธโฃ Biocontainment strategies for safely deploying C. acnes in human skin ๐๐ฆ  2๏ธโฃ A proof-of-concept therapy to combat UV-induced oxidative damage โ๏ธ๐ฅ

             Thus, we have focused in engineering C. acnes.
