6/6 This study highlights the important considerations for the design of future clinical trials combining cellular therapy with checkpoint inhibition and the complexity of this combination.

5/6 and activation. On the other hand, ipilimumab was associated with reduced donor NK cell persistence, suggesting that this was mediated by the rapid recovery of CD8+ T cells, rejecting the donor cells via HLA mismatch.

4/6 We conducted extensive correlative studies to evaluate the CIML NK cells after their infusion using flow cytometry and single cell CITEseq. CIML NK cells shifted towards early proliferative and maturation. Ipilimumab was associated with an increase in CIML proliferation.

3/6 under the hypothesis that this will lead to depletion of Tregs, via ADCC. Overall, treatment of CIML NK cells with N-803 was safe, with only 1 patient experiencing a DLT. Though responses were modest, there was a correlation between NK cell expansion and tumor reduction.

2/6 We describe the clinical outcomes of 10 patients who received CIML NK cells from a related haploidentical donor, and treated with an IL-15 superagonist (N-803) to expand and activate them in vivo. The trial was later amended to add the checkpoint inhibitor ipilimumab,.

1/6 Our Manuscript presents the clinical outcomes and correlative studies of our trial using cytokine-induced memory-like (CIML) NK cells for the treatment of advanced head and neck cancer.
