Modern GWAS can identify 1000s of hits but it can be hard to turn this into biological insight. What key cellular functions link genetic variation to disease? I'm excited to present our new work combining associations + Perturb-seq to build interpretable causal graphs! A🧵

1/ We report @ImmunityCP that the PD-1:PD-L1 axis promotes the development of B-cell memory and antibody responses in mice and humans. To our surprise, the pro-B-cell effect of PD-1 and PD-L1 is, at least partially, mediated in a B-cell-intrinsic manner. https://t.co/bUQetIShiF

Excited to share our new paper on temporal analysis of vaccine+/- adjuvant hours to 100 days (14 time pts). Vax alone induces new baseline immune state early & persists to 100 days, and that state predicts vax responses in an indep cohort. https://t.co/r59HePuN1t.

So many great institutions and people supported me over those years. The best part completing my thesis was writing the acknowledgments. Thank you all... @SidneySussex @MedCambridge @NIAIDTraining @DrAimeeLee @hollern_daniel @DrChrisHourigan @ChenZhao2011 @rohitfarmer

I'm honored to be recognized among my peers in @NIHOxCam. The program's unlimited flexibility + lack of boundaries allowed me to focus on learning & follow the science wherever it led. So grateful to @TsangLab, Ken, @plyons34 & many others for guidance https://t.co/NJZ4XI4n6y 1/2

Imagine a patient at initial presentation with an immune-mediated disease; rather than focusing on tissue/antibody-based diagnosis, you treat based on a test that captures the patient's global immune health. Another great step in that direction from my colleagues @tsanglab

1/12 Check out this paper that came out last week from our pan-monogenic project, which utilizes diverse natural single-gene defects in humans to study and assess the immune health of both patients with disease or those clinically healthy. https://t.co/4VOsJMCgIc

There's been amazing coverage of our new paper @Nature https://t.co/qkrUksBYVW but I wanted to put together a walk-through (and make this accessible as possible). It's a bit long so get a cup of tea! The full version is open access so pls do have a read if you are interested...

..individual immune systems illustrated as leaves...past encounters with caterpillars shape each leaf like the exposures and environmental inputs that shape immune systems. Vividly colored leaves represent individuals who exhibit a shared "naturally adjuvanted" immune setpoint.

It's so meaningful to share this chapter of work with my colleagues @TsangLab with my wife Sarah, who created the cover for @ImmunityCP. Days spent working together while in grad / art school, overlooking our garden in Cambridge, were some of the happiest in my life :) 1/

Using spatial metabolic flux we identified the conditions to preserve discarded human donor kidney’s for at least 4 days ex vivo. Such an approach also holds the potential for resuscitation and could increase the window for organ allocation. https://t.co/gN27A13npx

Our paper @ImmunityCP reveals a human baseline setpoint state (ie, "temperature" under which immune responses are initiated) linked to better responses to flu vaccination resembles cell states induced early and uniquely by the AS03 adjuvant. 1/n
