Cells & organoids within hydrogels can remodel their surroundings through pericellular/peri-organoid ECM secretion. Thus, our system, could map the dynamics of pericellular matrix remodeling, and be harnessed to create tissues for regen med. (20/23).

We also incorporate the FRET sensor into hydrogels using a cysteine’s thiol group. This should allow for its use in a range of synthetic and biologically derived hydrogels or cell-derived ECMs (Matrigel, e.g.) with little to no additional modification. (19/23)

The enzyme cleavage site we used can also be replaced by sequences recognized by specific MMPs. So, the platform can be tailored to look at (e.g.) MMP9 for studying inflammatory bowel diseases or MMP3 for looking at branching morphogenesis in models of development. (18/23)
