Importantly, sEcad remodels the brain microenvironment by promoting reactive astrocytes through the CXCL1–CXCR2 axis; blocking this pathway reduced brain metastasis burden and prolonged survival.

Using novel IBC brain metastasis models, we show that sEcad drives brain metastasis by impacting multiple steps of the metastatic cascade, including invasion, anoikis resistance, endothelial adhesion, and BBB transmigration.

We describe novel tumor cell-intrinsic and - extrinsic mechanisms by which soluble E-cadherin (sEcad) promotes brain metastatic growth in aggressive breast cancers. Serum sEcad levels are correlated with increased development of brain metastasis and death in IBC patients.
