This work stems from @GrilloAnthonyS’s idea to compile over a decade worth of data in these mice. We are grateful for the help of countless undergraduate students who gathered all the data, and for @mkaeberlein who funded the majority of the studies these data are taken from. 6/n.

Thus, these mice can be used to test longevity interventions in a mammal in a short period of time. Also, this model demonstrates that geroscience can be successfully applied to non-age-related pathologies and bring new perspectives in our understanding of chronic disease. 5/n.

Other longevity interventions, such as low O2, NMN, and AKG are also effective, which raises the question of why treatments with different targets are successful in this model. We argue that aging and mitochondrial disease have shared underpinnings in the “Hallmarks of Aging” 4/n.

Ndufs4-/- mice live on average for about 2 months and have characteristic brain pathology and metabolic abnormalities. Remarkably, longevity interventions such as rapamycin, acarbose, and 17-alpha estradiol increase survival in this mouse model. 3/n.

Ndufs4-/- mice lack a key component of Complex I of the Electron Transport Chain and are thus incapable of mitochondrial respiration. They model a pediatric mitochondrial disease called Leigh Syndrome, a neurometabolic disorder that is currently incurable and poorly managed. 2/n.

Help save the Dog Aging Project. Despite being one of the most influential, productive and impactful NIH supported projects over the past 5 years, the National Institute on Aging has inexplicably chosen to withdraw support for the Dog Aging Project. For now, I’m won't get into.
