Longer-lived mammals have evolved under different selection pressures ๐๐ถโโ๏ธ๐ถ. Complex mammalian tissues face unique challenges and must maintain homeostasis over months and years (if you're a lab mouse) and even decades (humans). So variation is an expected feature, not a bug.

So - what does all this mean in the wider context? Well, there is a lot of evidence that autophagy pathways generally decrease over the lifespan in different models throughout life ๐ชฐ๐ชฑ๐งซ. We think this is true. To be clear, we have no issue with this assertion. However... 7/n

We hope our map of mitophagy and macroautophagy in the aging brain will prove of high utility to researchers in both academia and industry who are actively working on the therapeutic modulation of these pathways, and have intentions to target the CNS. 6/n

We also profiled mitophagy and macroautophagy in different neuronal populations in vivo (e.g. A9, A10 DA neurons) and regional niches of astrocytes + microglia and also the CSF-producing choroid plexus of the aging brain... but one of the most interesting surprises was inhibitory

Pexophagy is the new mitophagy! So happy โ HIF1 upregulates NIX, which independently localises to mitochondria and peroxisomes to mediate their autophagy. Great work from @LeaPwilhelm and our collaborators in the labs of @patricia_boya and @AshToye_Bristol
