Most importantly, we learned that CDS (unlike Ewing sarcoma) can probably be modeled in mice, however, innovative approaches to prevent spontaneous recombination will be required. Stay tuned!.

So what did we learn? Well, for starters, CIC-DUX4 is a very potent oncogene which seems to behave as a neomorphic transcriptional activator in line with prior findings.

As it turns out, the problem wasn’t just leaky expression of the inserted fusion gene but spontaneous loxP recombination; a rare event in Cre-LoxP models that normally goes unnoticed.

Sort of. Unfortunately, all of the tumors developed in our chimeric animals which had never been exposed to Cre recombinase. Worse yet, the burden of disease was so extensive that nearly all of the animals died before breeding age.

Amazingly, all of the mice developed aggressive small round cell tumors which grew rapidly and metastasized early mimicking the disease course (and the appearance) of human CDS. So that’s good, right?
