🧠 New Research on Sleep-Wake Patterns and Alzheimer's! 💤.We found frequent, irregular interruptions of daytime activity and nighttime sleep were associated with higher levels of Alzheimer's plaques in the brain. #Alzheimer #SleepResearch #EndALZ.

Fragmented 24-hour activity rhythms were associated with higher subsequent amyloid-β burden, suggesting that rhythm disturbances have preceded amyloid-β deposition and may be a modifiable risk factor for Alzheimer disease.

💪This is promising first evidence that we may be able to transport AD prediction models from the research to the primary care setting - an important step for the successful application of new AD treatments.

🔍 Importantly, these findings were validated in the prospective population-based Rotterdam Study.(n = 500) with higher accuracy (e.g., AUC increased from 0.73 to 0.85), suggesting high generalisability to a population with very different characteristics than the training sample.

📊 Our best performing model included age, APOE ε4 genotype, family history of dementia, and subjective/objective measures of cognition, walking duration, and sleep. With an AUC of 0.73, predictive performance was moderate in the training and internal test set (A4 Study, n=4,119).

💡Our goal was to determine how accurately prediction models, developed in a large convenience (clinical) sample with a wide range of easily ascertainable predictors, could identify amyloid-positive individuals from a population-based sample.

Very proud to share our first amyloid-PET publication in the population-based Rotterdam Study where we showed that diabetes is associated with increased amyloid-beta pathology seven years later, independent of genetic dementia risk #ENDALZ @ErasmusMC .

Important work lead by @frankjwolters that shows, we need more population-based studies in dementia research. Probably plasma biomarkers are an important step in this direction #ENDALZ @ErasmusMC.

We found a strong relationship between vascular risk factors and cerebrovascular disease. Yet, education was NOT a strong modulator of this relationship.
