Interestingly, performing single cell RNA seq analysis we observed a higher frequency of geographically clustered gene repression across somatic chromosomes in clusters that emerge after senescence stress stimulus.

Given the spatial dimensions of the observed DNA dense and transcriptionally silent globules we reasoned that associated transcriptional effects operate on a scale larger than any individual gene—that is, repression of chromosomal segments.

We applied multi-isotope imaging mass spectrometry (MIMS)-method of nanoscale quantitative imaging of stable isotope tracers, to quantitatively map newly synthesized RNA in senescent cells. We identified hypercondensed, transcriptionally silent DNA globules in senescence.
