In contrast, the elongation rates of the various formins were much less impactful on ring assembly, in agreement with the computational model. We also looked at the effect of formin mechanoinhibition on ring assembly– Check it out!

By engineering yeast strains that target different formins to the cytokinesis nodes in place of WT formin Cdc12, we observed robust ring assembly for formins having similar nucleation properties to Cdc12 and defects for formins with dissimilar nucleation.

We explored how changes to these properties impact cytokinetic ring formation in fission yeast. Our simulations indicated that formin's ability to nucleate filaments was significantly more impactful than the rate at which it elongates them.

Formins shape the cytoskeleton by nucleating and elongating actin filaments. Within a single cell, multiple formin isoforms are targeted to specific actin networks, and each of them have distinct nucleation and elongation properties.

We present ~2.8 A cryo-EM structures of actin alone and nucleotide-free (rigor) wild-type myosin-15 bound to F-actin, as well as wild-type myosin-15 in the ADP state (3.6 A) and the jordan mutant the rigor state (3.8 A) to investigate how myosin-15 regulates actin assembly.
