smol-seq can be run in parallel to any other sequence-based method like RNAseq, opening up easy spatial metabolomics or single-cell metabolomics. We believe that this is a true nextgen metabolomics platform that can transform how we detect metabolites and drugs.

The SSAs are exquisitely specific — they can distinguish between very similar molecules (eg glucose vs galactose, ATP vs GTP etc) but even between stereoisomers. Since each SSA has a different barcode readout we can read out many thousands of them in parallel.

A glucose sensor releases one barcode, a cortisol sensor releases a different barcode. By sequencing released barcodes, we can quantify which metabolites are present and can read thousands of sensors in parallel. Metabolomics thus becomes a simple problem of DNA sequencing.

smol-seq relies on structure-switching aptamers (SSAs) to detect metabolites and give a DNA sequence output. How does this work? SSAs go through a large conformational change when they bind their target. We use that change to drive the release of a short unique DNA ‘barcode’.

#BREAKING: A pickup truck crashes into a TTC bus at high speed at Yonge/Finch leaving 8 people injured including driver of pickup who ran off but was arrested. The 7 people hurt on the bus include the bus driver. None of the injuries are life threatening.

@riken_en Finally, this was a very satisfying project as it all began as basic science: throwing drugs on worms and seeing what happened, with no disease relevance in mind. That led us to a new assay, a new drug screen, a new family of leads. Basic science opens doors!!
