I’m looking for a PhD-level scientist to join my research group @Vir_Biotech. We work on exploratory projects in molecular virology and virus-host interactions across multiple viral pathogens with the goal to uncover new therapeutic approaches.

Great to see this story finally out. It was started with my first research associate @czbiohub Ashley Ngo, who brought immense enthusiasm and dedication to the lab every day.

Knockout or knockdown of these restriction factors greatly enhanced the number of AAV-transduced cells and transgene expression levels. We hope that these insights may contribute to new strategies to avoid viral vector silencing and thus enable more efficient gene therapies.

We revealed components of the DNA damage response incl. the Fanconi Anemia pathway. We also found chromatin+transcriptional regulators, e.g. the histone methyltransferase SETDB1, which famously silences retroviral sequences, and MORC3 shown to restrict herpesvirus infection.

The presence of anti-AAV NAb or capsid-directed cytotoxic T cells are well known. By contrast, the cell intrinsic barriers for optimal transgene expression are not fully understood. We performed CRISPR screens to analyze restriction factors that inhibit AAV transgene expression.
