This was further supported with ALI organoids, in which pre-incubation with IFNg was sufficient to limit Crypto’s ability to replicate. Thanks as ever to everyone at Penn for your help and support - looking forward to following up on this in the Pardy lab!!.

By treating IFNg KO mice with IFNg, we showed that it took 24h to see a reduction in parasite burden. Crypto’s life cycle lasts 12h so this suggests IFNg acts on uninfected cells to limit dissemination within the host.

Some highlights: We used single-cell RNA seq of intestinal epithelial cells to identify IFNg-responsive cells that emerged during infection in the middle to tip of the villus, which correlated with parasite localization.
