Protrudin sits within an interacting network of proteins implicated in endosomal tubule fission, meaning it is ideally placed to co-ordinate elements involved in this process.

- finally, the endosomal tubule fission process is enriched for proteins involved in motor neuron disorders, and we show that human cortical neurons lacking protrudin or KIF5 develop abnormal endosomal tubulation, consistent with a fission defect.

-this means the process is mechanistically (and likely spatially) distinct from protrudin's known role in late endosome motility, which requires FYCO1 - results are consistent with a model where protrudin facilitates a tug-of-war between KIF5 and dynein to fission the tubules

- these phenotypes can be rescued by wild-type protrudin, but not by forms that cannot bind VAPs at the ER or endosomal phosphoinositides, or by forms unable to bind KIF5 - KIF5 depletion causes a similar phenotype, but depletion of FYCO1 does not
