(5/6).Promoter-proximal tethering elements facilitate co-regulation across a vertebrate-like promoter-promoter distance. Deletion of a single element (in green) maintains the proximal gene’s activity, but the distal gene never turns on in 1/3 of the nuclei.

(4/6) .What underlies co-regulation?. CRISPR manipulations identify:. A shared enhancer underlying the activities of both genes in an anterior domain. Promoter-proximal elements, distinct from the enhancer, that allow for long-range tuning of transcription dynamics.

(2/6) .Simultaneous live imaging of transcription from two distant genes in living Drosophila embryos. Imaging in an anterior stripe domain of 2-3h embryos shows concurrent transcription of knirps (green) and knirps-related (red), in single nuclei (blue).
