Our new paper is out:.Tumors don’t just reprogram monocytes locally. We show in a mouse model that the systemic impact of a tumor can alter the epigenome of peripheral monocytes, repressing IFN-responsive genomic sites and priming pro-tumor gene programs.

We also detected epigenetic priming in peripheral monocytes at promoters of genes previously linked to tumor-promoting functions (e.g. Trem2, Cd38, Thbs1, Myc).

This epigenomic programming affected functionally important genes. For example, promoters/enhancers that are responsive to immunostimulatory interferons (harboring IRF or ISRE TF binding motifs), were repressed genome-wide in monocytes already before their arrival to the tumor.

We could link these epigenetic changes in peripheral monocytes to sustained induction/repression of neighbouring genes, which persisted after tumor infiltration. We also observed epigenetic priming in peripheral monocytes at genes that were activated only later in the tumor.

By analyzing various histone modifications (H3K4me3, H3K4me1, H3K27ac) and chromatin accessibility genome-wide, we found that peripheral monocytes in tumor-bearing hosts show extensive epigenetic alterations at their promoters and enhancers.
