Our study demonstrates that the metabolic role of ADO in clearing cysteamine is important for cancer cells. Loss of ADO led to polyamine enrichment, which caused upregulation of PRODH, stimulating proline catabolism and mitochondrial activity. This in turn led to toxic ROS levels

The ADO knockout mouse taught us that ADO plays a role in embryonic development but is dispensable in adult murine tissues – at least in the absence of stress. In contrast, ADO is needed in cancer cells to support their proliferation, survival and growth as xenografts. Why?!.
