In summary, we show that a pOXA-48-encoded regulator mediates a plasmid-chromosome transcriptional crosstalk, resulting in a growth benefit for the plasmid-carrying bacterium ➡️ This could promote plasmid maintenance and transmission in clinical strains.

Finally, by expressing the pOXA-48-encoded lysR in trans and by silencing the pfp-ifp operon using CRISPRi technology, we demonstrated that the overexpression of the pfp-ifp operon is beneficial for pOXA-48-carrying Klebsiella pneumoniae - 9/10 (10/10 👇)

Backed up with functional annotations of the PFP and IFP proteins and our transcriptomic and experimental results, we hypothesized that the overexpression of the pfp-ifp operon in the presence of pOXA-48 could lead to an increased iron uptake - 8/10

We also performed transcriptomic analyses of strains carrying the lysR deletion mutant of pOXA-48, and demonstrated that the LysR-mediated transcriptional crosstalk is specific to the pfp-ifp operon genes - 7/10

pOXA-48 encodes a LysR regulator that is highly similar to the LysR that controls the pfp-ifp operon. Through genetically engineering a pOXA-48 lysR deletion mutant, we demonstrated that the pOXA-48-encoded LysR is responsible for the overexpression of the pfp-ifp operon - 6/10

Remarkably, pOXA-48 induced the overexpression of a small chromosomal operon (formed by the pfp and ifp genes) in Klebsiella spp. and Citrobacter freundii. Interestingly, the pfp-ifp operon, along with its lysR regulator, shows signs of being horizontally mobilizable - 5/10

We analyzed the transcriptional response of 11 MDR clinical enterobacteria to carriage of the carbapenem-resistance plasmid pOXA-48. First, we observed that the most expressed plasmid gene was the carbapenemase blaOXA-48 - 3/10
