Hence, we suggest that not only cell-cell rearrangements, but also cell-cell communication are likely to work synergistically and biochemical and mechanical processes evolve under a common morphogenetic timescale in the system.

But is that all? what about the difference in the timing of symmetry breaking depending on the initial fraction of T+ cells? Our proportion experiments can be explained assuming that pluripotent stem cells (A) inhibit mesoderm differentiation of Bra+ cells (B).

Could there be mechanical differences between Bra+ and Bra- tissues leading to cell sorting? The answer was yes! By combining fusion experiments and nanoindentation we found Bra+ tissues to have a higher surface tension than Bra- tissues. 🧐

Those aggregates as well as gastruloids followed the same progression: At 24h, Nanog+ (magenta) and Bra+ (green) were found in a salt and pepper configuration, while around 48h islands of T- cells (Aldh1a2, Gata6 in yellow) formed on the outside, surrounding a core of Bra+ cells

Heterogeneity on the initial levels of T expression is known to influence gastruloid formation. We decided to form aggregates with controlled proportions of T+ and T- cells and found that increasing the initial T+ cell population speeds up polarization and elongation.

We are excited to share the latest contribution from the lab! We found that cells in gastruloids communicate to each other in order to control the timing of gastruloid symmetry breaking.
