https://t.co/jaz7IUE2mC Congrats to Dr. Sunita Keshari and Dr. Alex Shavkunov for putting together a nice story that contributes to our understanding of neoantigen cancer vaccines and different immune checkpoint therapies. @kesharisunita @AlspachLab @KenHu77 @kepauken @kchenken

The goal of therapeutic cancer vaccines and immune checkpoint therapy (ICT) is to eliminate cancer by expanding and/or sustaining intratumoral T cells with enhanced anti-tumor capabilities. However,...

I've been advocating for greater transparency around UAP for over a decade. Now, SENATOR Chuck Schumer and Senator Rounds, in a bipartisan fashion, are pushing through and unprecedented Bill that will bring it all out into the light. HOUSE Intel and armed services are trying
House Republicans are trying to kill the Senate's bipartisan measure to increase transparency around UAPS. The measure I’m championing with Sen. Rounds would create a board to work through the declassification of government records on UAPs. We'll keep working to get this done.

Are so-called 'M2-like' CD206+ macrophages immunosuppressive? On the contrary, we find that they are critical for T cell mediated anti-tumor immunity https://t.co/XFOe4dknmL with @MaxKrummel @KenHu77 @KellyKersten8 @alexis_combes and others

Thanks for the shoutout @Simon_J_Cleary . Was a real pleasure working with you and Francis! It's difficult to keep abreast of the fast moving field of spatial transcriptomics, but here's my brief take on its intersection with immunology.
Francis Man & I edited a book! Here are some shout-outs to contributors & highlights of ‘Imaging Inflammation’! https://t.co/wI6Vc1d3ms

A neat feature of examining gene programs is identification of conserved gene programs across datasets based on correlated gene weightings. We used this to probe the idea of the "tumor as the wound that never heals" in mouse tumor models and patient samples.

We verified some of these spatiotemporal patterns using imaging of cleared whole wounds or thick sections of wounds. CE3D worked brilliantly to preserve endogenous fluorescence while also allowing antibody staining. https://t.co/dJ5UmeyICN

By examining correlation of these patterns we could identify cell cell interactions, and importantly the downstream target genes induced by the signal in the receiver cells (e.g. between fibroblasts and macrophages)

We can then plot gene expression level, cell type frequencies and gene program levels as a function of both dimensions:

As with many dynamic multicellular processes, the axes of space and time are essential. We sampled the wound at varying space time coordinates for scRNA-Seq of the CD45+ and CD45- compartments
