Out today @J_Immunol from first authors @bhesh82 & Mohi Choudhury: .✅NLRP3-dependent cell death occurs in CASP1- & GSDMD-deficient cells with LPS + ATP, suggesting cell death pathway other than pyroptosis.✅Led us to identify NLRP3-PANoptosome in WT cells.

The team found that NLRC5 was critical for pathology across inflammatory diseases, including hemolytic disease, #colitis, and #HLH models. The findings suggest targeting NLRC5, or other molecules in this pathway, has therapeutic potential in hemolytic and inflammatory #disease.

Heme release is often associated with hemolytic diseases (like #sicklecelldisease & #malaria), but many infectious & inflammatory diseases, including #SARSCoV2 & others, cause red blood cells to break apart and spill their contents, releasing heme to activate innate immunity.

NLRC5 then went on to induce the formation of a PANoptosome complex that contained an NLR network, with NLRP12 and the famous #NLRP3 also present. This led to inflammatory cell death.

This study found new functions for an enigmatic innate immune sensor, NLRC5. In response to heme and specific components of #infection (PAMPs) or cellular damage (#TNF), as well as depletion of the energy storage molecule NAD+, NLRC5 expression was upregulated.
