I'm excited to share my lab's first preprint! It describes Affinity Map, a highly general method providing proteomic profiles of binding affinity for small molecules, peptides, and antibodies in cell lysate, organ extracts, and live cell surfaces.

Because µMap labeling works equally well for all types of protein targets, Affinity Map can be applied in challenging biological settings (live cells, cell lysate, brain membrane extract) and is compatible with many types of ligand (small molecules, peptides, proteins).

Our approach is similar to a radioligand competition assay, where the affinities of both the "hot" (photocataltyic) and "cold" (unmodified) ligands are simultaneously measured by measuring labeling while systematically varying the concentrations of both molecules.

Affinity Map combines high throughput proteomics, plate-based µMap labeling workflows that use an improved, super-hydrophilic photocatalyst, and classic Cheng-Prusoff competitive binding kinetics to provide an accurate apparent binding affinity (Kd) for every detected protein.

Amazing to see a plan come together! This paper unambiguously shows the tight labeling radius of antibody-targeted µMap labeling compared to other methods, and how reactive intermediate lifetime can be exploited to tune proximity labeling radii.

Our TIMSTof Pro 2 (a cutting edge LCMS for proteomics) is now installed and fully operational! Here is a picture of Alan, the Bruker rep, manuvering it with help from Maxim and Zahir. It extends vertically into the ceiling, as all awesome instruments should.
