The other big surprise: as we used µMap on HER2-low cancers, we kept finding Protein Tyrosine Phosphatase F (PTPRF) as a pan-cancer HER2 interactor. Turns out its broadly upregulated in cancer and knockdown suppresses proliferation in HER2-low cancers.

We found that galectins were uniquely enriched in trastuzumab-resistant models. Inhibiting galectins genetically or pharmacologically actually restore d sensitivity through destabilization of HER2 and enhancing antibody-mediated degradation!.

Using our high-res photocatalytic µMap proximity labeling platform, we comprehensively profiled the HER2 interactome across human breast cancer lines, allowing us to pinpoint novel mechanisms of HER2 dependency & resistance to targeted therapies like trastuzumab.
