Given the co-expression of TrkB and mGluR5 in the nervous system and their roles in synaptic plasticity. The enhancement of TrkB-driven synaptic plasticity by mGluR5 positive allosteric modulators offers promising avenues for advanced antidepressant treatments.

1. The identified crosstalk between TrkB and mGluR5 is facilitated through the cooperative action of Gbg, released from TrkB, and Gaq-GTP, released from mGluR5. This represents a novel and physiologically significant form of RTK/GPCR crosstalk.
