We ran our first official DNA gel in the beautiful shared space of the Miller Lab and @dafang07 Lab. Many many thanks to @taraenders for helping my student and I with the 20 things I forgot I needed to run a gel. Time to order more things! Glow-up lab reno pics coming soon!

This was such a fun project to work on, and a fantastic team of scientists to do it with (including my first undergraduate mentee to be an author on a paper with me!). Genetic screens are my jam, and I relish any opportunity to show all of the great discoveries they can lead to.

What's more! Again, with the skills of @therickybaker, we were able to identify possible structural similarities between our new activation mechanism and how Ral GTPases activate the exocyst in animal cells.

What does this mean? The exocyst is not a simple machine (duh!) that simply switches between "on/off" or "open/closed". Rather that there are multiple ways to activate it - and each results in discrete, insulated outcomes.

This second activation mechanism which is mediated by an interaction between the tomosyn homolog Sro7 and the exocyst subunit Exo84 is distinct from Cdc42/Rho activation of the exocyst.

Super excited to share that our newest paper is out @JCellBiol ! https://t.co/HHZ2PfBK3b - In this multidisciplinary effort with the Brennwald lab and @therickybaker we identify a new allosteric activation mechanism for the exocyst! A short 𧡠on some of the highlights:
