News from @AmelioLab: Advanced #SalivaryCancer and developing glands share common #p53 pathway regulation. Pro-tumorigenic FGFR2 distinguishes foetal development from high-grade MEC. https://t.co/8y4XpwKvMj

4. Finally, we used computational and molecular analyses of human and murine salivary MEC tumors to reveal that alternative splicing of FGFR2 distinguishes C1/M2-positive MEC tumors from transcriptional programs operational in normal fetal development.

3. We developed an autochthonous mouse model of salivary MEC in which expression of the C1/M2 oncogene is targeted to Krt14-positive ductal cells in the murine salivary gland and confirmed that C1/M2 expression coupled with Tp53 loss drives progression to high-grade MEC.

2. While C1/M2 fusion positive salivary MECs generally harbor wild-type p53, we observed progressive elevation in MDM2 expression levels according to tumor grade suggesting that post-translational loss of p53 contributes to disease progression.

1. We performed a comprehensive integrated analysis of gene expression data, which revealed that activation of developmental signaling programs is a hallmark of high-grade salivary MEC. These analyses also revealed common p53 pathway regulation in development and tumorigenesis.

While low-grade MEC tumors can be successfully managed with surgical resection, patients with high-grade tumors have limited treatment options. Thus, understanding the signaling mechanisms involved in progression is crucial for identifying novel therapeutic vulnerabilities.

Mucoepidermoid carcinoma (MEC) is the most frequently occurring salivary gland malignancy. This tumor type is characterized by a t(11;19) chromosomal translocation, which generates the oncogenic fusion CRTC1-MAML2 (C1/M2).

Thrilled to share our story out in Cancer Cell, led by the amazing Kay Hanggi (now on the job market). We show that necrotic forms of cell death and the subsequent release of IL1a drives myeloid-based immune suppression in models of breast cancer. (1/11) https://t.co/7cafJW2KSQ
